α-klotho and Alzheimer’s disease –new article-

α-klotho has been known as a protein involved in aging and our bestselling product #27998 Human soluble α-Klotho Assay Kit – IBL has been widely used for research in bone metabolism and chronic kidney disease (CKD).

In recent study, it has been reported that α-klotho involves in Alzheimer’s disease (AD) in several scientific articles.

The following recent article published by a study group of University of California, San Francisco (UCSF) who are supported by several public institutes such as National Institute on Aging (NIA) in April 2017 is available online.

The functional haplotype of klotho gene “KL-VS” is a variant mutated F352V and C370S that it has been considered that it influences for function of klotho protein secretion.

Serum klotho levels were increased with KL-VS heterozygosity while it was decreased with KL-VS homozygosity in this study.

It is suggested that elevated klotho levels promote a resilient brain possibly through increased network connectivity of critical brain regions.

Systemic klotho is associated with KLOTHO variation and predicts intrinsic cortical connectivity in healthy human aging. Yokoyama JS et al. Brain Imaging Behav. 2017 Apr;11(2):391-400. PMID: 27714549

The research group led by Johns Hopkins University School of Medicine, National Institute on Aging (NIA) and National Institutes of Health (NIH) also released an interesting their study result that indicates klotho levels in CSF of older adults with Alzheimer’s disease (AD) was much lower than older adults without AD and klotho levels in CSF was higher in men than women and higher in young normal adults than elderly people.

About Alpha klotho (α-klotho)

“α-klotho” was identified as an extremely down-regulated gene in the genetically-modified mouse of which phenotype is very similar to various symptoms of human aging.

Then, the sequences of α-klotho genes have been identified in various species including humans based on the one of mouse. α-klotho protein is a 130 kDa, one-transmembrane protein and its expression is confirmed in the kidneys and the parathyroid glands. In recent years, it has become clear thatα-klotho is an important molecule within a living organism regulating the metabolism of mineral such as calcium and phosphorus.
