Q.Can it be measured in urine sample?

A.Yes, it is measurable, but it is unstable.
The dilution ratio should be evaluated and determined by each laboratory, but it is considered as it might be in the range between 10,000 and 20,000 fold. In this case, the commercially available product specific dilution buffer (Code: 27799D-30mL) is additionally required.

Q.Does Neuramidase treatment require for the sample?

A.No it doesn't. There is no much different for the result between with and without Neuramidase.

Q.I found the recommended dilution ratio is between 200 and 800 fold. That is quite a wide range. What is the most optimized dilution ratio?
-
A.Some samples have a very high value of Gd-IgA1 and such samples would be range over that why the recommended dilution ratio is wide.
We recommend to start from 200 fold dilution and find the best dilution ratio for your samples. The optimized dilution ratio should be determined by each user. -
Q.What is approx. normal IgA concentration in blood?
-
A.Total IgA is approx 1-4mg/mL in blood.

Q.What is approx. normal Gd-IgA1 concentration in blood?
-
A.As it is approx 1,000 ng/mL in blood, more than 200 fold dilution is recommended for measurement.

Q.What is the dilution ratio for urine sample?

Q.What is the condition of DPP 4 inhibitor?

A.Only EDTA-plasma collected using collecting tubes contained DPP4 inhibitor can be used for measuring of GLP-1 active form. BD™ P800 GLP-1, GIP, Glucagon, Ghrelin storing vacuumed collecting tube should be used for collecting blood or the blood should be blocked its activity by DPP4 inhibitor prior it is measured. The amount of DPP4 inhibitor for use is 10μL per 1mL blood or 20μL per 1mL plasma.

Q.This kit can be used for measurement of diabetes drug (e.g. Liragrutide)?

A.As this kit was developed to measure native GLP-1 active form (7-36/37), we have not evaluated the kit with any commercially available drugs.

Q.What are the epitopes of pre-coated antibody (Anti-mouse Klotho 162A1 Rat IgG) and HRP conjugated antibody (Anti-mouse Klotho 235A1 Rat IgG Fab')?

A.Klotho has two domains (KL1 and KL2). Although we have not conducted epitope mapping, these antibodies can detect KL1 domains.
The kit was tested using culture supernatants expressing only KL1 of mouse Klotho and has shown a positive response. -

Q.How to wash an ELISA plate?

A.Washing it by an auto-washer is highly recommended.

Q.The wells turned black during the test with the kit.

A.It is possible that the wells were not washed sufficiently during the washing process after the HRP-labeled antibody reaction.
Be sure to wash the wells enough times as described in the data sheet with washing buffer of more than 350 µL.

Q.What is the condition of DPP 4 inhibitor?

A.Only EDTA-plasma collected using collecting tubes contained DPP4 inhibitor can be used for measuring of GLP-1 active form. BD™ P800 GLP-1, GIP, Glucagon, Ghrelin storing vacuumed collecting tube should be used for collecting blood or the blood should be blocked its activity by DPP4 inhibitor prior it is measured. The amount of DPP4 inhibitor for use is 10μL per 1mL blood or 20μL per 1mL plasma.
